Description

Solis Hot Start HiFi Master Mix is a 5x-concentrated, ready-to-use solution designed for precise singleplex and multiplex PCR applications. It features Solis Hot Start HiFi DNA Polymerase with 3′→5′ exonuclease (proofreading) activity, offering over 100x higher fidelity compared to wild-type Taq polymerase, as demonstrated through single-molecule real-time (SMRT) sequencing.

The polymerase in the master mix is chemically modified to enable hot-start PCR, enhancing specificity and accuracy, minimizing mispriming and extension from non-specifically annealed primers and primer-dimers. This makes the mix suitable for applications that demand high fidelity PCR, such as sequencing, cloning, and long-range PCR – supporting amplification of fragments up to 22 kb DNA depending on the target).

With improved tolerance for both GC- and AT-rich regions (27–79% GC content), the mix ensures reliable amplification of challenging targets. Its superior multiplexing performance enables even amplification of diverse amplicons, making it ideal for complex assay designs.

The DNA polymerase included in Solis Hot Start HiFi Master Mix has efficient toleration of dUTPs, making it well suited for amplification of bisulfite treated DNA (e.g., for bisulfite sequencing), damaged DNA (e.g., FFPE samples). Robust performance is further supported by demonstrated inhibitor tolerance, ensuring consistent and reliable results even in the presence of common PCR inhibitors.

Additionally, the Solis Hot Start HiFi Master Mix is room temperature stable for up to 1 month due to the use of Stability TAG – Solis BioDyne’s proprietary and patented polypeptide stabilization technology that is incorporated into the enzyme, enabling environmentally friendly ambient temperature shipping [1]. Room temperature stability also enables maximum convenience allowing reactions to be set up without ice at room temperature.
