Overview

Description

FIREScript® FLEX H(-) Reverse Transcriptase (RT) is an in silico engineered RNA-directed DNA polymerase that can synthesize a complementary DNA strand from ssRNA or ssDNA. It is designed for high-sensitivity cDNA synthesis and robust performance under challenging conditions.

The enzyme operates efficiently across an extended temperature range (37°C-70°C) and contains reduced RNase H activity, preserving RNA templates and enabling accurate full-length cDNA synthesis. Its high processivity supports extended transcript coverage, including long and complex RNA templates.

FIREScript® FLEX H(-) RT demonstrates exceptional thermostability, maintaining activity after prolonged heat exposure (65°C for 30 minutes) with minimal impact on Ct values (see the figure below). In addition, the enzyme exhibits enhanced benchtop stability, delivering consistent performance even after reaction setup is held at room temperature for extended periods, helping to reduce variability and improve reproducibility. It has enhanced stability at room temperature as well as 37°C with no activity loss for up to 1 month due to the use of Stability TAG – Solis BioDyne’s proprietary and patented polypeptide stabilization technology that is incorporated into the enzyme, enabling environmentally friendly ambient temperature shipping [1]. In addition, the enzyme supports benchtop workflow robustness, remaining stable at room temperature for up to 24 hours during reaction setup, enabled by its inherent warm-start properties. Room temperature stability also enables maximum convenience allowing reactions to be set up without ice at room temperature.

This combination of thermal robustness, RNA preservation, and high processivity makes FIREScript® FLEX H(−) RT particularly well suited for demanding applications, including low-input, degraded, or structurally complex RNA.

For optimal performance, the use of RiboGrip® RNase Inhibitor is strongly recommended.
