Description

SOLIScript® Lyo-ready 1-step RT-qPCR Kit is an optimized solution for fast and sensitive amplification and quantitation of RNA and DNA targets with probe-based assays. With a high collapse temperature (Tc) of -30 ˚C, the kit allows for the adoption of a rapid and energy-efficient lyophilization protocol. With a glass transition temperature (Tg) of 85.9˚C, the lyophilizates have a high resistance against long term exposure to higher temperatures. The composition is optimized for lyophilization to cakes or beads. The kit comes in 4 tubes: 40x SOLIScript® Lyo-compatible One-Step Mix, HOT SolisFAST® Glycerol-Free DNA Polymerase (10 U/μl), 4x Lyo Reaction Buffer and 4x RT-qPCR Lyo Excipient Mix.

40x SOLIScript® Lyo-compatible One-Step Mix is a 40x concentrated RT mix that includes an in silico designed SOLIScript® reverse transcriptase and RiboGrip® RNase Inhibitor. SOLIScript® is a thermostable reverse transcriptase active at temperatures up to 60°C, beneficial when using templates with high levels of secondary structure. RiboGrip® inactivates RNase A to protect RNA sample from degradation and increase cDNA yield in reactions with low RNA amounts.

HOT SolisFAST® Glycerol-Free DNA Polymerase (10 U/μl) is an in silico designed inhibitor tolerant analogue of Taq DNA polymerase with chemical hot-start and ~2-4 times faster extension rates compared to the wild-type Taq.

4x Lyo Reaction Buffer contains additives for efficient amplification. The MgCl2 concentration is 20 mM (1x mix – 5 mM).

4x RT-qPCR Lyo Excipient Mix is a 4x concentrated blend of lyophilization additives that protect reagents during freeze-drying and stabilize the lyophilizates. The mix enables lyophilization into both cakes and beads.

Yes, it is possible to opt for your own set of lyophilization excipients with the SOLIScript® Lyo-ready 1-step RT-qPCR Kit. However, for the most optimal lyophilization conditions and to achieve the most stable lyophilizates, we highly recommend using the dedicated 4x RT-qPCR Lyo Excipient Mix.

What are lyophilization excipients?

To protect the biological material, such as enzymes, from the temperature and pressure changes during lyophilization, freeze-drying protectants are employed. These protecting excipients ensure that the materials reserve their active state after lyophilisation. In addition to that, they provide an adequate structure to the lyophilized cake or bead.

What is lyophilization?

Lyophilization, commonly known as freeze-drying, is a process employed to remove the water content from a substance by sublimation after it has been frozen, while preserving its structure and properties. The reagents can be lyophilized to a spheric lyo-bead or a compact lyo-cake format.

Lyophilization helps to extend the product’s shelf life, enhance the stability of the reagents at higher tempereatures, even when mixed together with primers and probes, and allows for convenient and accurate dispensing of the reagents. After lyophilization, the reagents are easily reconstituted with sample material.
